I Scooby-Doos största äventyr hittills får vi för första gången se hur de oskiljaktiga Scooby och Shaggy lärde känna varandra och hur de tillsammans med de unga detektiverna Fred, Velma och Daphne grundade Mysteriegänget. Efter att ha löst hundratals fall ställs Scooby och gänget inför sitt största och svåraste mysterium: en ondskefull komplott för att släppa lös spökhunden Kerberos. När de kämpar för att stoppa den globala "hundpokalypsen" upptäcker gänget att Scooby har ett hemligt arv och ett storslaget öde, större än någon kunnat vänta sig.
